Welcome to the Winogradsky column lab page! Students from the Departments of Biological Applications and Technology, University of Ioannina and Icthyology and Aquatic Environment, University of Thessaly, Greece and the Microbiology course, Faculty of Sciences, University of Cádiz, Spain, discuss their findings on Winogradsky columns they constructed!

Univ. of Thessaly, Group number 04 (Ev. Gousia - Chr. Gkizas), Day 0-50, Incubation: Dark

We are undergraduate students in the Department of Ichthyology and Aquatic Environment, Agriculture Science, University of Thessaly, Volos, Greece. We are presenting our results from the Winogradsky column experiment that was conducted in Anauros beach (Volos) (21/03/2016). We colected results of  50 days .

When the sample collected at the first day, into the bottle there were one broken egg, pieces of paper and sediment (brown colour) and water (light yellow colour) of the area.

 
After 7 days of the experiment we 've noticed that the sediment became black and a white substance appear in the top of it. The water colour turned to ligth brown.

After 17 days the sediment was black and the water became white. The thick, white substance at the top of the sediment has disappeared.

After 30 days the water became dark gray and a gray substance appeared above the sediment.

After 36 days no changes at the column.

After 50 days the sediment became more compact and black, and the colour of the water became dark gray.

Our names are Theano Stamopoulou and George Sountoulidis and we are undergraduate students in the Department of Ichthyology and Aquatic Environment, Agriculture Science, University of Thessaly, Volos, Greece. We are presenting our results from the Winogradsky column experiment that was conducted in at Anauros beach (Volos) (21/03/2016). We colected results of 53 days .


The first day the sediment had a ligth brown color and the water was ligth yellow

After 7 days of our experiment we 've noticed the first significant changes in our column. The sediment became darker. The water in the bottle turned into white and a white substance was created at the top of the sediment.

After 20 days the water in the bottle turned into light grey (first picture) and a white substance was created in the surface (second picture). The white substance at the top of the sediment has disapeared.

 After 30 days the sediment became darker (first picture)  and the substance at the top of the bottle was thicker and whiter (second picture).

After 40 days : We didn't notice any changes.

After 53 days only the colour of the sediment became darker.

UCA_1A-9ª_1: Day 70. Our last day in the column.

First of all, our dark column is getting darker, that means sulfate-reducing bacterias keep growing! Secondly, we have noticed that water is cleaner than las time. We think that's because there were some bacerias in it but they have died and gone down.

In the light column, the green zone is bigger because the algae keep growing too! Nevertheless, red marks are similar as las time, so it's probably that iron-reducing bacterias have died.

We lit a lightner near the top of the bottle to know if some bacerias had produced methane but the test was negative in both column.

UCA_8A_4: Day 72 – Finally, some changes appeared!

Rio San Pedro sediment + 0.5g FeSO4 + filter paper
As you can see in  the picture our column is still the same. The black stains of FeS still at the bottom and there are a big amount of them.

However, this time some little orange stains appeared at the surface because of the FeO2 synthezised by the bacteria. That means the Fe2+ reacted with oxygen and formed those orange stains. There's still more water than the last time.

Volos_Group 9_Light

Our names are Ilianna Kaltsatou and Ioanna Kalogiri and we are undergraduate students Department of Ichthyology and Aquatic Environment, University of Thessaly, Volos, Greece.
I'm presenting my results from the Winogradsky column experiment that was conducted in 1/4/2016 at Anauros beach (Volos) since today 26/5/2016.

They not observed excessive fluctuations, in terms of sediment color, turbidity of the water, etc.

Today,on day 45(25/5/2016).We are presenting you our results from day 20 to day 65


Day 40

The odor of our experiment its worse than  the last time due to the presence of hydrogen sulfide (H2S).The lower layer of the sediment its much more darker due to the presence of organic matter.The colour of the water its white-yellow and its different than the last time.

Here we can see that the colour of the water its grey and its different than the last time (DAY 65).Also,the water level its lower than the last time.






















We can observe that the colour of the water on the surface layer is white-grey and that means that the microorganismos are dying.

Hi everyone! We have observed both colums after other two weeks.




If you look up and down the bottle to which it has affected him the light, we can see that the water bottle is murkier due to the presence of cyanobacteria. Also at the top we are more amount of O2




If we go down in the bottle, we can distinguish three colors. These colors are green, yellow, orange and bright red. The color green is found near water and is due to the presence of cyanobacteria, which are producing O2


In the area of ​​yellow color, we find sulfur oxidizing bacteria, which use carbon as a source of organic matter. Find oxygen (bubbles) and hydrogen sulfide. Orange areas is due to the presence of red non-sulfur bacteria, in the presence of organic carbon source gives it color. The shades of light red color is due to the presence of red sulfur bacteria. In this area we find O2 as they use as reducing the H2S and generate sulfates.






The last area is black. In this area reducing bacteria sulfate. Using the sulfate product gives as H2S which react with the Fe (II) to produce iron sulfide, which is precipitated and this color giving the last zone.







 Comparing the column to light with the column to the dark can see that the water is too murky. We observed a thin layer of orange due to oxidation of iron. The color of the bottle is gray due to organic matter. Near the bottom of the bottle we look dark colors. By not meet the light, we will not see big changes in the bottle. Here hardly we find microorganisms.









 The bottle on the left is the one about which the light has influenced. However, the bottle on the right is the one about which the darkness has influenced
